CIMmultus® Monoliths are reusable chromatography devices for scalable high-resolution purification of complex biological samples. Inside the custom designed housing is a single piece stationary phase with homogeneous channel size and surface chemistry. This stationary phase is designed to simplify method transfer from development into production.
They can be used as single-use disposable or multi-use columns, and they are available in various volume ranges – from 1mL for screening and method development purposes up to 40L for industrial manufacturing.
The strongly hydrophobic structure of polystyrene divinylbenzene (SDVB) enables reversed-phase purification of nucleic acids and other biomolecules.
This high-performance column is especially useful for separating between double-stranded and single-stranded species of similar length. CIMmultus® SDVB gives its best results as a polishing method. It can be used for fast, low pressure and room temperature purification of RNA, including RNA exceeding 10 kb (such as self-amplifying RNA).
CIMmultus® SDVB is commonly but not exclusively used with an ion-pair reagent. The ion-pair reagent neutralises the negative charge on the RNA backbone to improve binding of RNA or other nucleic acid to the monolith. Elution is achieved by increasing concentration of organic solvent such as acetonitrile.
SDVB (Poly-styrene divinylbenzene) features:
- Strongly hydrophobic surface for RP or ion-pair RP chromatography
- Enables size separation and purification of nucleic acids (DNA, RNA)
- High caustic stability, flow independent performance, ease of scalability
- Available as CIMac analytical column for HPLC analytics
Benefits:
- Very fast processes by convective mass transport – resolution independent of flow
- Ready to use columns
- High-capacity for large molecules due to fully accessible void volume in the channels
- Laminar flow minimizes shear stress resulting in high yields
