Overview

Specifications

CIMmic® Oligo dT is intended for capturing of ssRNA with a poly-adenylated tail, making in an ideal tool for process development screening of chromatographic conditions for your mRNA. CIMmic Oligo dT combines the accessibility of the CIMmic format with oligo-based affinity chemistry, providing a simple and flexible tool for small-scale purification and process development of mRNA.

The ligand in CIMmic® Oligo dT18 is an 18-mer deoxythymidine chain covalently immobilised on the monolith with a carbon linker. RNA with a poly(A) tail binds to the ligand by hybridisation between adenine (A) bases on the RNA tail and deoxythymine (T) bases of the ligand. Species that do not contain a poly(A) tail do not bind to the column and are found in the unbound (flow-through) fraction.

Oligo dT (deoxythymine) features:

  • Oligo dT ligands covalently bound to a solid support
  • Hybridises to the poly-adenylated tail which is present on most eukaryotic mRNAs, or synthetised onto the molecule during IVT
  • Primarily intended for fast and efficient capture and purification of mRNA with a poly-adenylated tail, from various sources
  • Can selectively remove impurities from the sample
  • Convenient approach for initial purification of mRNA

Benefits:

  • Fewer unit operations: apply IVT material to monolith without buffer exchange
  • Very fast processes by convective mass transport – resolution independent of flow
  • High-capacity for large molecules due to fully accessible void volume in the channels
  • Laminar flow minimizes shear stress resulting in high yields
  • Ideal for early process development and feasibility testing
  • Operable by syringe or FPLC
  • Low sample and buffer consumption
