CIMmultus® Monoliths are reusable chromatography devices for scalable high-resolution purification of complex biological samples. Inside the custom designed housing is a single piece stationary phase with homogeneous channel size and surface chemistry. This stationary phase is designed to simplify method transfer from development into production.
They can be used as single-use disposable or multi-use columns, and they are available in various volume ranges – from 1mL for screening and method development purposes up to 40L for industrial manufacturing.
The ligand in CIMmultus® Oligo dT18 is an 18-mer deoxythymidine chain covalently immobilised on the monolith with a carbon linker. RNA with a poly(A) tail binds on the ligand by hybridisation between adenine (A) bases on the RNA tail and deoxythymine (T) bases of the ligand. Species that do not contain a poly(A) tail do not bind on the column and are found in the unbound (flow-through) fraction.
Oligo dT (deoxythymine) features:
- Oligo dT ligands covalently bound to a solid support
- Hybridises to the poly-adenylated tail which is present on most eukaryotic mRNAs, or synthetised onto the molecule during IVT
- Primarily intended for fast and efficient capture and purification of mRNA with a poly-adenylated tail, from various sources
- Can selectively remove impurities from the sample
- Convenient approach for initial purification of mRNA
